Abstract

Official abstract text for this publication.

This present invention relates to an improved process to prepare prostacyclin derivatives. One embodiment provides for an improved process to convert benzindene triol to treprostinil via salts of treprostinil and to purify treprostinil.

First claim

What is claimed is: 1. A method of preparing a pharmaceutical product from a pharmaceutical batch that can be stored as a stable compound at ambient temperature and that is prepared by a method comprising (a) alkylating a benzindene triol, (b) hydrolyzing the product of step (a) to form a solution comprising treprostinil, (c) contacting the solution comprising treprostinil from step (b) with a base to form a salt of treprostinil, wherein purity is at least 99.5% as determined by HPLC area under the curve analysis, comprising storing the pharmaceutical batch at ambient temperature and preparing a pharmaceutical product from the pharmaceutical batch after storage. 2. A method as claimed in claim 1 , wherein the salt of treprostinil is a diethanolamine salt. 3. A method of preparing a pharmaceutical batch of a salt of treprostinil that can be stored as a stable compound at ambient temperature and that is prepared by a method comprising (a) alkylating a benzindene triol, (b) hydrolyzing the product of step (a) to form a solution comprising treprostinil, (c) contacting the solution comprising treprostinil from step (b) with a base to form a salt of treprostinil, wherein purity of the batch is at least 99.5% as determined by HPLC area under the curve analysis, comprising (a) alkylating a benzindene triol, (b) hydrolyzing the product of step (a) to form a solution comprising treprostinil, (c) contacting the solution comprising treprostinil from step (b) with a base to form a pharmaceutical batch of a salt of treprostinil and d) storing the pharmaceutical batch at ambient temperature. 4. A method as claimed in claim 3 , wherein the salt of treprostinil is a diethanolamine salt.
