The present invention relates to a class of monoclonal antibody that specifically binds the phosphorylated serine 396 residue on pathological hyperphosphorylated (PHF) tau (pS396) with improved affinity, as well as to methods of using these molecules and their tau binding fragments in the treatment of Alzheimer's disease and other tauopathies.

What is claimed is: 1. A monoclonal antibody comprising: (a) a Light Chain CDR1 having the amino acid sequence of SEQ ID NO:31; (b) a Light Chain CDR2 having the amino acid sequence of SEQ ID NO:4; (c) a Light Chain CDR3 having the amino acid sequence of SEQ ID NO:5; (d) a Heavy Chain CDR1 having the amino acid sequence of SEQ ID NO:6; (e) a Heavy Chain CDR2 having the amino acid sequence of SEQ ID NO:7; and (f) a Heavy Chain CDR3 having the amino acid sequence of SEQ ID NO:8. 2. The monoclonal antibody according to claim 1 , further comprising: (a) a Light Chain comprising the amino acid sequence of SEQ ID NO:16; and (b) a Heavy Chain comprising the amino acid sequence of SEQ ID NO:11. 3. A pharmaceutical composition comprising the monoclonal antibody according to claim 1 and a pharmaceutically acceptable carrier.
