Embodiments herein report compositions, uses and manufacturing of dengue virus constructs and live attenuated dengue viruses. Some embodiments concern a composition that includes, but is not limited to, a tetravalent dengue virus composition. In certain embodiments, compositions can include constructs of one or more serotypes of dengue virus, such as dengue-1 (DEN-1) virus, dengue-2 (DEN-2) virus, dengue-3 (DEN-3) or dengue-4 (DEN-4) virus constructs. In other embodiments, constructs disclosed herein can be combined in a composition to generate a vaccine against more one or more dengue virus constructs that may or may not be subsequently passaged in mammalian cells.

What is claimed: 1. A nucleic acid sequence comprising a nucleic acid sequence encoding a modified live, attenuated dengue-2 virus represented by SEQ ID NO:13 (WVS). 2. A pharmaceutical composition comprising the nucleic acid sequence according to claim 1 , and a pharmaceutically acceptable excipient. 3. A method for inducing an immune response in a subject comprising, administering the pharmaceutical composition of claim 2 to the subject, wherein the composition induces an immune response in the subject against at least one dengue virus serotype. 4. A vector comprising a nucleic acid sequence according to claim 1 . 5. The pharmaceutical composition according to claim 2 , further comprising one or more live, attenuated dengue viruses having a nucleotide sequence encoding a dengue virus or dengue virus chimera represented by SEQ ID NOS: 1, 3, 5, and 7, which is a dengue-1/dengue-2 chimera. 6. The pharmaceutical composition according to claim 2 , wherein the composition contains at least one, live attenuated dengue virus that represents each of the four dengue virus serotypes, wherein the composition is a tetravalent composition. 7. The pharmaceutical composition according to claim 2 , further comprising an immunogenic composition comprising a live, attenuated flavivirus selected from the group consisting of yellow fever virus, tick-borne encephalitis virus, Japanese encephalitis virus, West Nile virus, hepatitis C virus and a combination of two or more thereof. 8. A method for inducing an immune response in a subject comprising, administering the pharmaceutical composition of claim 5 to the subject, wherein the composition induces an immune response in the subject against at least one dengue virus serotype. 9. The method composition according to claim 8 , wherein the composition contains at least one, live attenuated dengue virus that represents each of the four dengue virus serotypes, wherein the composition is a tetravalent composition. 10. A kit comprising the immunogenic composition according to claim 5 , and a container. 11. A dengue-2 nucleic acid sequence represented by SEQ ID NO: 13 encoding species containing nucleotide G or C at nucleotide position 6481. 12. A dengue-2 nucleic acid sequence represented by SEQ ID NO:13 encoding species containing nucleotide C or T at nucleotide position 7156. 13. An isolated cell comprising at least one of the nucleic acid sequences according to claim 1 . 14. A method for manufacturing a pharmaceutical composition comprising, combining at least one of the nucleic acid sequences according to claim 1 and a pharmaceutically acceptable excipient and making a pharmaceutically acceptable composition for use in inducing an immune response against dengue virus in a subject.
