This invention is in the area of tricyclic lactam compounds for and methods of treating selected Rb-positive cancers and other Rb-positive abnormal cellular proliferative disorders while minimizing the deleterious effects on healthy cells, for example healthy Hematopoietic Stem Cells and Progenitor Cells (HSPCs), associated with current treatment modalities. In one aspect, treatment of select Rb-positive cancers is disclosed using specific compounds disclosed herein. In certain embodiments, the compounds described herein act as selective cyclin-dependent kinase 4/6 (CDK 4/6) inhibitors when administered to subjects.

We claim: 1. A method for treating retinoblastoma (Rb)-positive breast cancer in a host comprising administering an effective amount to a host in need thereof, of a compound selected from: or a pharmaceutically acceptable salt thereof. 2. The method of claim 1 , wherein the host is a human. 3. The method of claim 1 , wherein the compound is administered in combination with another chemotherapeutic agent. 4. The method of claim 3 , wherein the chemotherapeutic agent does not rely on cellular proliferation for its anti-cancer activity. 5. The method of claim 2 , wherein the compound is or a pharmaceutically acceptable salt thereof. 6. The method of claim 5 , wherein the compound is administered in combination with another chemotherapeutic agent. 7. The method of claim 2 , wherein the compound is or a pharmaceutically acceptable salt thereof. 8. The method of claim 7 , wherein the compound is administered in combination with another chemotherapeutic agent. 9. The method of claim 2 , wherein the compound is or a pharmaceutically acceptable salt thereof. 10. The method of claim 9 , wherein the compound is administered in combination with another chemotherapeutic agent.
