The present invention provides for concentrated aqueous silk fibroin solutions and an all-aqueous mode for preparation of concentrated aqueous fibroin solutions that avoids the use of organic solvents, direct additives, or harsh chemicals. The invention further provides for the use of these solutions in production of materials, e.g., fibers, films, foams, meshes, scaffolds and hydrogels.

What is claimed is: 1. A composition consisting essentially of at least 4% wt silk fibroin protein but not more than 10% wt silk fibroin protein, water, and a therapeutic agent. 2. The composition of claim 1 , wherein said silk fibroin protein is obtained from a solution of dissolved silkworm silk, genetically engineered silk, or dissolved spider silk. 3. The composition of claim 1 , wherein the therapeutic agent is a small molecule, a protein, a peptide, a polysaccharide, a glycoprotein, a lipoprotein, or a nucleic acid. 4. The composition of claim 1 , wherein the therapeutic agent is a small molecule selected from the group consisting of vitamins, sedatives, hypnotics, antibiotics, antiviral agents, steroids, growth factor inhibitors, growth factor promoters, prostaglandins and radiopharmaceuticals. 5. The composition of claim 1 , wherein the therapeutic agent is selected from the group consisting of anti-infectives, chemotherapeutic agents, anti-rejection agents, analgesics and analgesic combinations, anti-inflammatory agents, hormones, anti-angiogenic agents, anti-thrombotics, anti-metabolics, antimitotics, fibrinolytics, and growth factors. 6. The composition of claim 5 , wherein the growth factor is selected from the group consisting of bone morphogenic proteins, bone morphogenic-like proteins, epidermal growth factor, fibroblast growth factor, platelet derived growth factor, insulin-like growth factor, transforming growth factors, and vascular endothelial growth factor. 7. The composition of claim 1 , wherein the therapeutic agent is an antibody. 8. The composition of claim 1 , wherein the therapeutic agent is present as a liquid or a finely divided solid. 9. The composition of claim 1 , wherein the therapeutic agent further comprises a pharmaceutically acceptable carrier. 10. A composition consisting essentially of at least 4% wt silk fibroin protein but not more than 10% wt silk fibroin protein, water, and an additive. 11. The composition of claim 10 , wherein said silk fibroin protein is obtained from a solution of dissolved silkworm silk, genetically engineered silk, or dissolved spider silk. 12. The composition of claim 10 , wherein the additive is a biologically and/or pharmaceutically active compound. 13. The composition of claim 10 , wherein the additive is an antibody, a deoxyribonucleic acid, a ribonucleic acid, a modified ribonucleic acid and protein composite, a glycogen, a sugar, an alcohol, or cytokine. 14. The composition of claim 10 , wherein the additive is a cell attachment mediator, a biologically active ligand, a growth factor, or an osteoinductive substance. 15. The composition of claim 14 , wherein the cell attachment mediator is selected from the group consisting of collagen, elastin, fibronectin, vitronectin, laminin, proteoglycans, a peptide containing known integrin binding domains and a combination thereof. 16. The composition of claim 14 , wherein the growth factor is selected from the group consisting of fibroblast growth factor, epidermal growth factor, platelet-derived growth factor, insulin-like growth factor, transforming growth factor, and a combination thereof. 17. The composition of claim 14 , wherein the osteoinductive substance is a bone morphogenic protein. 18. The composition of claim 10 , wherein the additive is a bioactive substance that functions as a receptor or chemoattractant for a desired population of cells.
