This invention relates macrocyclic purine derivatives, processes for their preparation, pharmaceutical compositions, and their use in treating viral infections.

First claim

The invention claimed is: 1. A compound having formula (I) or a pharmaceutically accepted salt thereof, wherein X is oxygen, NH, sulfur or Y is phenyl or pyridinyl, optionally substituted by one or more substituents independently selected from C 1-6 alkyl, C 1-4 alkoxy, trifluoromethyl or halogen, and Z is C 1-10 saturated or unsaturated alkyl optionally substituted by an alkyl or alkylhydroxyl; or Z is C 1-6 alkyl —NH—C(O)—C 1-6 alkyl- or C 1-6 alkyl —NH—C(O—C 1-6 alkyl —O—; or Z is C 1-10 alkyl —O— wherein said alkyl is unsaturated or saturated and can optionally be substituted by an alkyl or alkylhydroxyl; or Z is C 1-6 alkyl-O—C 1-6 alkyl- wherein said alkyl is unsaturated or saturated and can optionally be substituted by an alkyl or alkylhydroxyl; or Z is C 1-6 alkyl-O—C 1-6 alkyl-O— wherein said alkyl is unsaturated or saturated and can optionally be substituted by an alkyl or alkylhydroxyl. 2. A compound selected from the group consisting of: or a pharmaceutically acceptable salts thereof. 3. A pharmaceutical composition comprising a compound having formula (I) or a pharmaceutically acceptable salt thereof according to claim 1 and one or more pharmaceutically acceptable excipients, diluents or carriers. 4. A method of activating TLR7 in a subject comprising administering to the subject a therapeutically effective amount of a compound as claimed in claim 1 . 5. A pharmaceutical composition comprising a compound according to claim 2 and at least one pharmaceutically acceptable excipient, diluent or carrier. 6. A method of activating TLR7 in a subject comprising administering to the subject a therapeutically effective amount of a compound according to claim 2 . 7. A method of inducing interferon in a subject comprising administering to the subject a therapeutically effective amount of a compound of claim 1 . 8. A method of inducing interferon in a subject comprising administering to the subject a therapeutically effective amount of a compound of claim 2 .
