Compounds and methods of using the same to inhibit glycolysis and treat cancer and other diseases are disclosed.

What is claimed is: 1. A method of treating brain cancer or pancreatic cancer in a patient in need thereof comprising administering to the patient a therapeutically effective amount of one or more compounds of the Formula I: or a salt, ester or prodrug thereof, wherein: each R 1 , R 2 , R 3 and R 4 are independently H, or C(O)R 5 ; at least two of R 1 , R 2 , R 3 and R 4 is C(O)R 5 ; and each R 5 is independently selected from C 3 -C 7 straight- or branched-chain alkyl, C 4 -C 10 alkylcycloalkyl, and C 3 -C 7 cycloalkyl. 2. The method of claim 1 , wherein R 2 and R 3 are C(O)R 5 . 3. The method of claim 1 , wherein the compound is chosen from: or a salt thereof. 4. The method of claim 1 , wherein the cancer treated is pancreatic cancer. 5. The method of claim 1 , wherein the cancer treated is brain cancer. 6. The method of claim 5 , wherein the brain cancer is selected from the group consisting of glioblastoma, high-grade glioma, and metastatic brain cancer. 7. A method of treating brain cancer or pancreatic cancer in a patient in need thereof comprising administering to the patient a therapeutically effective amount of the compound or a salt thereof. 8. A method of treating brain cancer or pancreatic cancer in a patient in need thereof comprising administering to the patient a therapeutically effective amount of the compound or a salt thereof.
