Provided herein are (alpha-substituted aralkylamino or heteroarylalkylamino) pyrimidinyl and 1,3,5-triazinyl benzimidazoles, e.g., a compound of Formula I, and their pharmaceutical compositions, preparation, and use as agents or drugs for treating proliferative diseases.

What is claimed is: 1. A method for the treatment or amelioration of one or more symptoms of a PI3K mediated cancer in a subject, which comprises administering to the subject a compound of Formula I: or an enantiomer, a mixture of enantiomers, a mixture of two or more diastereomers, or an isotopic variant thereof; or a pharmaceutically acceptable salt, solvate, hydrate, or prodrug thereof; wherein: X, Y, and Z are each independently N or CR X , with the proviso that at least two of X, Y, and Z are nitrogen atoms; where R X is hydrogen or C 1-6 alkyl; R 1 and R 2 are each independently (a) hydrogen, cyano, halo, or nitro; (b) C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 3-10 cycloalkyl, C 6-14 aryl, C 7-15 aralkyl, heteroaryl, or heterocyclyl;
