The present disclosure provides antibodies that bind to interleukin-3 receptor alpha chain and uses thereof.

First claim

The invention claimed is: 1. A method for inducing lysis of leukemic cells in a subject with acute myeloid leukemia (AML), the method comprising administering to a subject suffering from AML an isolated or recombinant antibody that is capable of specifically binding to interleukin (IL)-3Rα chain, wherein the antibody comprises: (i) a light chain variable region (V L ) comprising CDRs 1, 2 and 3 as set forth in SEQ ID NOs: 2, 3 and 4, respectively; (ii) a heavy chain variable region (V H ) comprising CDRs 1, 2 and 3 as set forth in SEQ ID NOs: 5, 6 and 7, respectively; and (iii) a heavy chain constant region comprising amino acid substitutions S239D and I332E according to the EU numbering system of Kabat. 2. The method of claim 1 , wherein the antibody is a humanized antibody comprising: (i) a light chain variable region (V L ) comprising an amino acid sequence according to SEQ ID NO: 8 and a heavy chain variable region (V H ) comprising an amino acid sequence according to SEQ ID NO: 9; and (ii) a heavy chain constant region comprising amino acid substitutions S239D and I332E according to the EU numbering system of Kabat. 3. The method of claim 1 , wherein the heavy chain constant region comprises a sequence set forth between residues 121-450 (inclusive) of SEQ ID NO: 11. 4. The method of claim 1 , wherein the antibody is a humanized antibody comprising a light chain comprising a sequence set forth in SEQ ID NO: 13 and a heavy chain comprising a sequence set forth in SEQ ID NO: 11.
